計畫中文摘要	本計畫結合了電化學及營養物選殖生物刺激技術，進行含氯有機物之整治效能評估與分析。本研究的重點包括：(1)以菌群代謝預測分析技術，記錄三氯乙烯(TCE)降解過程的生態位。(2)藉由改變含TCE汙染物樣本之電子供體及受體提供率，提升微生物降解TCE汙染物之速率。本研究中作為生物刺激添加劑的泥火山樣本菌群代謝功能分析結果顯示，最常見之代謝機制是化學異營及硫化物暗氧化反應。葡萄糖類降解較少的泥火山樣本，其甲烷氧化相關代謝功能的菌群相對較高。而Ecoplate分析結果可作為樣本的碳源添加依據。利用KEGG數據庫進行甲烷氧化和硫代謝相關的潛在酶的特定基因分析結果，亦為泥火山樣本之硫代謝和甲烷氧化基因的存在提供驗證。泥火山樣本之Spearman相關性分析結果顯示，許多硫還原細菌與甲烷氧化相應的基因pmoA、pmoB和pmoC呈顯著正相關。在pH和ORP較高的條件下，甲烷氧化菌能更有效率進行甲烷氧化。在本研究之微生物燃料電池(MFC)模組試驗中發現，Dehalococcoides mccartyi和Dehalobacter一旦在MFC陰極定殖，可以加速對TCE進行還原脫氯。添加泥火山灰的MFC樣本組，會增加甲烷氧化和甲基營養代謝路徑。這兩種代謝路徑對TCE降解具有正面影響。在三種不同類型之MFC模組中，以含火山灰的固-液相MFC模組其TCE代謝效率最佳。
計畫英文摘要	In this study, a well-designed experimental framework was established to integrate electrochemistry and biostimulation for effectively exploring the degradation efficiency of chlorinated organic compounds. The experimental setup encompasses the following key components: (1) Ecological Niche Exploration in Trichloroethylene (TCE) Degradation Process: A comprehensive investigation was conducted into the ecological niche associated with the TCE degradation process. This involved a combination of microplate medium utilization and an analysis of projected microbiota metabolism. Insights into the ecological conditions conducive to TCE degradation were gained through the analysis of microbiota metabolic activity. (2) Microbial Degradation Rate Assessment: The overall rates of TCE degradation enhancement were quantified through controlled manipulation of electron donor and acceptor supply rates. The metabolic functionalities within mud volcano samples reveal various mechanisms, with chemolithotrophy and anaerobic sulfide oxidation reactions emerging as the most prevalent. Furthermore, the presence of genes relevant to methane oxidation and sulfur metabolism in the mud volcano samples is confirmed through our analysis of specific genes associated with these processes, utilizing the KEGG database. The mud volcano samples with lower rates of glucose degradation exhibit a comparatively higher abundance of microbial communities associated with metabolic functions related to methane oxidation. The Ecoplate analysis findings provide a practical foundation for the introduction of carbon sources to the samples. A strong positive relationship between a multitude of sulfur-reducing bacteria and genes responsible for methane oxidation (pmoA, pmoB, and pmoC) is demonstrated by the Spearman correlation analysis. Additionally, methane-oxidizing bacteria are observed to function more efficiently under conditions of higher pH and ORP. Within our Microbial Fuel Cell (MFC) module experiments, the colonization of the MFC cathode by Dehalococcoides mccartyi and Dehalobacter accelerates the reductive dechlorination of TCE. Upon the incorporation of mud volcano samples into the MFC configuration, a noticeable increase in methane oxidation and the methylotrophic metabolic pathways is observed. Both of these pathways contribute positively to the dechlorination of TCE, underscoring their potential for environmental applications. Among the three distinct types of MFC modules tested, the solid-liquid phase MFC module containing mud volcanic ash emerges as the most efficient in TCE metabolism.
